Key Responsibilities:

The TDS Proteins Development API organization is seeking a candidate interested in protein process development. Release glycan analysis is a critical quality attribute for therapeutic antibodies to determine the glycosylation profile which can impact efficacy, safety and stability of the molecule. There are multiple techniques available to analyze glycans from sample preparation to detection. Glycans can be released enzymatically and analyzed via mass spectrometry for more detailed structural information using minimal sample amount. Alternatively, the released glycans can be fluorescently labeled and run using either hydrophilic interaction liquid chromatography (HILIC) releasing glycans based on polarity or by capillary electrophoresis; these techniques offer high-resolution separation and quantification. The current method in CEAS-Analytical is via reduced mass spectrometry while analytical development typically runs a HILIC method to quantify glycans. Sciex has introduced a release glycan kit for use on the Biophase which could prove useful for quantification of glycans. In this role, the Co-Op will...

  • Compare these techniques to examine the feasibility and benefit of adding a labeled release glycan analysis to the CEAS-Analytical workflow. This will impact the business by bridging the gap between CEAS and downstream process development for oligosaccharide quantification.
  • Analyze and interpret experimental results using analytical and statistical techniques. Actively participate in weekly meetings with internal and external collaborators.
  • Participate in technical discussions on design of experiments to fill gaps in knowledge.
  • Maintain a laboratory notebook and other technical documents according to required guidelines.
