Antibody binding triggered by vaccination with polysaccharide/protein antigens does not always translate into effective killing mediated by neutrophils or macrophages. This behavior may stem either from the specific in vitro assay conditions or from the phenotype of some bacterial strains, which may reflect unique mechanisms of immune evasion.
What will you do?
The main objective of this project is to systematically optimize in vitro assay conditions to gain deeper insights into these mechanisms using polyclonal antisera targeting various polysaccharide or protein antigens, either individually or in combination, in presence of cells and complement and using different assays readouts
What will you learn?
Binding techniques to look at antigen-specific antibody quantities using multiplex methodologies.
Handling of pathogenic bacteria and eukaryotic cell cultures
Assays to evaluate the binding/functionality of vaccine-induced antibodies
Non-technical skills: Ability to follow standard operating procedures and work with the team to ensure maintenance of equipment and reagents in accordance with GSK and EHS requirements, development of problem-solving skills.
